缢蛏(Sinonovacula constricta)生化遗传特征分析

摘要:
于2001年4月, 在浙江近海采集100个缢蛏样品, 采用聚丙烯酰胺垂直板不连续电泳方法, 对缢蛏足、水管、消化腺、外套膜、鳃等5种组织或器官进行了EST、ADH、MDH、IDH、ME、G6PDH、LDH、ALP、GDH、CAT、ALP、ACP、SOD、SDH、ATP、SUDH、GCDH、AMY、GAD、POD等20种同工酶的电泳研究, 并对各种酶的同工酶位点及酶谱表型进行了分析。结果表明, 所测组织中未检出CAT、ALP、GCDH、GAD;AMY和ACP只在消化腺中有表达;SUDH只在水管、鳃、消 化腺中表达;所检测的酶均不同程度地表现出了组织差异。

BIOCHEMICAL GENETIC ANALYSIS OF SINONOVACULA CONSTRICTA

Abstract:
Sinonovacula constricta is an important economical species along the coast of China. In recent years, it has suffered heavy decrease in its resources, and its wild stocks and genetic diversity have been threatened to some extent due to overfishing and changes of ecological environment. It is the time to investigate the biochemistry of Sinonovacula constricta. In April 2001, 100 samples of Sinonovacula constricta were collected from Ningbo coast of Zhejiang Province. 5 tissues and organs i.e. gill, digestive gland (hepatopancreas), mantle, foot muscle and siphon were fetched from each of the samples and tested at once or stored in super cryo-refrigerator (-70℃) for test later. Tissues and organs were homogenized in 0.05% mercaptoethanol buffer at ratio of 1∶3 (W/V) with ice-bath and centrifuged by 12×103 r/min for 30 minutes at 4℃. The supernatant of samples to be used for the experiments was divided and put into 1.5 ml eppendoff tube, stored at -70℃. Twenty kinds of isozymes, including Esterase (EST), Alcohol dehydrogenase (ADH), Malate dehydrogenase (MDH), Isocitrate dehydyogenase (IDH), Malic enzyme (ME), Glucose-6-phosphate dehydrogenase (G6PDH), Lactate dehydrogenase (LDH), Alkaline phosphatase (ALP), Glutamate dehydrogenase (GDH), Catalase (CAT), Acid phosphatase (ACP), Superoxide dismutase (SOD), Sorbitol dehydrogenase (SDH), Adenosine triphosphatase (ATPase), Succinic dehydrogenase (SUDH), Glucose dehydrogenase (GCDH),α-Amlyase (α-AMY), Formale dehydrogenase (FDH), Galactose dehydrogenase (GAD) and Peroxidase (POD) in 2 buffer systems (TC, pH=7.0; TG, pH=8.3) were examined by the vertical polyacrylamide gel electrophoresis and their expression in 5 tissue and organs were tested. And the loci, phenotypes of these isozymes were also studied. The result showed that Sinonovacula constricta possess abound and complete enzyme system, it not only take part in metabolism and adjust as the form of enzyme, but also represent tissue-specificity on actvities and phenotypes. Catalase, Alkaline phosphatase, Glucose de- hydrogenase, Galactose dehydrogenase were not found expressing in the any kinds of 5 tissues. The activities of α-Amlyase, Acid phosphatase could be detected only in digestive gland, Peroxidase ingill, digestive gland and Succinic dehydrogenase in gill, digestive gland and siphon, While the others have wide distribution in the 5 tissues. The isozymic phenotypes of Malic enzym, Glucose-6-phosphate dehydrogenase, Superoxide dismutase, Lactate dehydrogenase, Sorbitol dehydrogenase, Isocitrate dehydyogenase was not found difference in the 5 tissues, and that of Adenosine triphosphatase, Esterase, Malate dehydrogenase, Glutamate dehydrogenase have evident difference among certain tissues, but Alcohol dehydrogenase, Formale dehydrogenase exhibited an apparent tissue-specificity. The physiological significance inrelation to the tissue-specific distribution of the isozymes was discussed.
