中国对虾三倍体诱导的研究

摘要:
中国对虾(Penaeus chinensis)主要分布于我国北方沿海,是我国水产养殖业的主要养殖对象,具有极其重要的经济价值。随着养殖事业的发展和人民生活水平的提高,提供优良的对虾品种已势在必行,于是遗传育种的研究引起了人们极大的重视。国家把生物技术在对虾上的应用列为国家“七五”和“八五”攻关课题。 近年来海洋生物多倍体诱导引起了水产养殖界的广泛兴趣(楼允东,1984;吴宝玲等,1986;姜卫国等,1987;王子臣,1990;相建海等,1992; Stanley,1981; Johnstone,1985;Downing,1987; Gosling,1989; Cai et al,1991),主要原因是三倍体较二倍体的生长速度快,长出的个体也比二倍体大,可得到较高的经济效益;同时,多倍体的诱导方法对染色体调控、性别决定和人工雌核发育(蔡难儿等,1995)等的研究都具有重要的意义。中国对虾的多倍体诱导不论从理论上还是从应用上都具有积极的意义,是一条值得探索的改良对虾品种的良好途径。

TRIPLOIDY INDUCTION IN PENAEUS CHINENSIS

Abstract:
This paper reports results from cold shock and Cytochalasin B treatment experiments designed to induce triploidy in Chinese shrimp, P. orientalis, One of the most important commercial species in China. Three major factors influencing the rate of triploids and hatching were analysed respectively to seek for the most suitable treatment conditions. Ploidy was assessed by counting the number of polar bodies associated with early embryos and the number of chromosome in appendage-budding stage. Bect results were achicved with cold shock (at 4℃ for 30min administered 7 min after spawning) or with cytochalasin B at 0.25mg/L (administered 22 to 32 min after spawning ). The triploidy percentages were 70% by cold shock and 48% by cytochalasin B treatment. Orthogonal analysis indicated that hatching rates were affected significantly by duration and temperature (for treatment by cold shock) and by dosage and duration (for treatment with cytochalasin B).
