对虾白斑综合征病毒VP28基因在大肠杆菌中的表达

摘要:
在已构建重组质粒pGEM-vp28的基础上,通过NcoⅠ,HindⅢ双酶切质粒pGEM-vp28和表达载体质粒pET-30a,分别获得携带限制性内切酶粘性末端的vp28基因片段和表达载体质粒片段,两片段经T4DNA连接酶连接获得了重组表达质粒pET30a-vp28,将此重组质粒导入表达宿主菌Escherichia coli BL21中,用IFID诱导表达4h后,其表达产物在SDS-PAGE和Westernblot中的显示的特异性条带大小约27ku,与预期的大小相近。

Expression of envelope-protein VP28-gene from white spot syndrome virus(VSSV) in Escherichia coli
Abstract:
On the base of the comstructed recombinant plasmid Pgem-vp28, VP28-gene fragment and expression vector plasmid fragment with limited inner-cut enzyme viscidity ends were gained by using NcoI and Hind Ⅲ cut plasmid Pgem-vp28 and expression vector plasmid pET-30a. An expression plasmid pET30a-vp28 including VP28-gene was obtained by T4DNA ligation enzyme. SDS-PAGE and Westem Blot show that objective gen has expressed in E. coli BL21 and the product is about 27 ku that is close to the size in anticipation.
