人工养殖军曹鱼亲鱼不同组织氨基酸的分析

摘要:
采用生化的方法,对人工养殖军曹鱼繁殖期间的肌肉、肝脏及性腺氨基酸含量进行测定。结果表明:(1)雄性亲鱼从Ⅳ期发育到Ⅴ期的过程中,精巢和肝脏牛磺酸含量、精巢总氨基酸含量均呈显著递增(P<0.05);肌肉氨基酸总量呈显著下降趋势(P<0.05);而肝脏氨基酸总量则无显著性差异(P>0.05);(2)雌性亲鱼从Ⅳ期发育到Ⅴ期的过程中,肝脏组织中牛磺酸含量呈显著升高(P<0.05);卵巢组织无显著变化(P>0.05);氨基酸总量差异变化规律与雄性亲鱼相类似。(3)亲鱼总氨基酸含量、总必需氨基酸含量在不同组织间存在显著差异(P<0.05),表现为肌肉组织含量最高,其次是性腺组织,肝脏组织中最低。肌肉、肝脏及性腺氨基酸组成类似,含量较高的为谷氨酸、天冬氨酸和赖氨酸;含量较低的为组氨酸、蛋氨酸和半胱氨酸。(4) 发育处于同期性腺雌雄亲鱼,雄性亲鱼肌肉、性腺必需氨基酸总量和氨基酸总量显著高于雌性亲鱼(P<0.05),而肝脏必需氨基酸总量和氨基酸总量无显著性变化(P>0.05)。

Analysis of amino acid composition in different tissues of cultured cobia (Rachycentron canadum) broodstock

Abstract:
To identify the effect of amino acid composition on the reproductive performance of cultured cobia broodstock, biochemical methods were used to determine amino acid composition in muscle, liver and ovary of mature fish (maturation stage Ⅳ and Ⅴ). The results showed that (1) between stage IV and V, the content of taurine in the testis and liver, as well as the total amino acid content in the testis were significantly increased (P<0.05) in male breeders; the total amino acid content in the muscle showed a significant decrease (P<0.05); while no significant difference of the total amino acid content was found in the liver (P>0.05). (2) In the female broodstock, the content of taurine increased significantly in the liver (P<0.05), but not in the ovary (P>0.05) during the gonad development from stage IV to stage V. The variation of total amino acid content in females was similar to that of males. (3) The contents of total amino acids and total essential amino acids were significantly different (P<0.05) among different tissues, the highest content was found in the muscle and the lowest in the liver. The amino acid content in the muscle, liver and gonadal glands were similar, high levels of glutamic acid, aspartic acid, and lysine were detected, and the contents were low in histidine, methionine, and cysteine. (4) The total essential amino acids and total amino acids in the muscle and gonad of male broodstock were significantly higher than those of female broodstock (P<0.05), no significant difference was found in the total amount of essential amino acids and amino acids in the liver (P>0.05).
