海洋酸化对马氏珠母贝珍珠层形成的影响

摘要:
分析了酸化环境对马氏珠母贝珍珠层形成的影响。在温度为28℃, 盐度为31 的条件下, 分别在pH 8.2(对照组, CG)、pH 7.7(实验一组, E1)和pH 7.4(实验二组, E2)的海水中养殖马氏珠母贝, 观察贝壳珍珠层结晶形貌的变化, 并检测珍珠层形成相关的矿化基因(pif177、nacreinpearlin)在外套膜中央区(mantle center, MC)的表达。结果表明: (1)通过扫描电镜观察贝壳, 比较对照组与实验组贝壳晶体形貌变化, 发现E1 组贝壳珍珠层低倍镜下, 生长阶梯明显, 高倍镜下超微结构规则, 六边形棱角不明显,但边界清晰; 而E2 组出现板块边界不明显, 生长排列不规则等现象, 说明了海水酸化影响了马氏珠母贝的钙沉积, 引起珍珠层形貌特征的变化; (2)实验设定条件下, pif177、nacreinpearlin基因表达量整体上呈现随pH 降低而降低的变化趋势。其中, E1 组与CG 组间均无显著性差异(P>0.05), E2 组均较CG组显著下调(P<0.05)。

Effects of ocean acidification on nacre formation and expression of related genes in Pinctada martensii
Abstract:
The current study examined the impacts of ocean acidification of on the nacre mineralization process of Pinctada martensii. The experimental pH values of the seawater for breeding experiments were 8.2 (controlling group, CG), 7.7 (experimental group 1, E1) and 7.4 (experimental group 2, E2) with other experimental conditions (28℃, 31‰ salinity) constantly. We observed changes in nacre crystal morphology and examined the expression levels of nacre mineralization associated genes (pif177, nacrein and pearlin) in the mantle center (MC). The results showed that the growth ladder of E1 group nacre was obvious in low magnification, while in high magnification, the ultrastructure was regular. Despite the hexagon edges were not obvious, the border was clear. However, the E2 group had no obvious plate boundary; arranged irregularly, further illustrating the significant impact of seawater acidification on calcification and the nacre crystal morphology of P. martensii. Under the experimental conditions, the expression of pif177, nacrein and pearlin, decreased with decrease in pH. Among them, there was no significant difference between the groups E1 and CG group (P>0.05), while the gene expression levels in E2 group were significantly lower than those in CG group (P<0.05).
