温度和pH 对杂色鲍消化酶活力的影响

摘要:
采用酶学分析方法研究了温度和pH 对杂色鲍(Haliotis diversicolor Reeve)肝胰腺和消化道中胃蛋白酶、类胰蛋白酶、淀粉酶和纤维素酶4 种主要消化酶活力的影响。结果表明, 在设定的温度和pH范围内, 杂色鲍各消化酶的活力均随着温度或pH 的升高呈现先升后降的变化趋势。其中, 肝胰腺和消化道中胃蛋白酶的最适温度均为50℃, 适宜pH 分别为2.6~3.0、2.6~3.4; 类胰蛋白酶在肝胰腺中的最适温度为40℃、适宜pH 为8.6~9.0, 在消化道中的最适温度为50℃, 适宜pH 为8.2; 肝胰腺和消化道中淀粉酶的最适温度分别为40、30℃, 最适pH 分别为6.8、7.2; 纤维素酶在肝胰腺中的适宜温度为40~50℃, 最适pH 为5.2, 在消化道中的最适温度为40℃, 适宜pH 为4.8。在最适温度和pH 范围内, 肝胰腺中纤维素酶、淀粉酶活力均显著高于消化道中, 具有器官特异性, 而胃蛋白酶和类胰蛋白酶活力在肝胰腺和消化道中差异不显著, 无器官特异性。同种消化器官内, 各消化酶的活力也存在差异。

Effect of temperature and pH on the activity of digestive enzymes in Haliotis diversicolor Reeve
Abstract:
The effects of reaction temperature and pH on the activities of 4 main digestive enzymes(pepsin, trypsin, amylase, cellulase) from hepatopancreas and digestive tract of Haliotis diversicolor Reeve were studied using enzymology analysis method. The results showed that the digestive enzyme activities of H. diversicolor Reeve were all first increased and then decreased with increased temperature or pH in the setted temperature and pH range. The optimum temperature of pepsin in hepatopancreas and digestive tract was both 50℃, the optimum pH range was 2.6~3.0 and 2.6~3.4 respectively. The optimum temperature of trypsin in hepatopancreas was 40℃ and the optimum pH was 8.6~9.0. The optimum temperature of trypsin in digestive tract was 50℃, and the optimum pH was 8.2. The optimum temperature of amylase in hepatopancreas and digestive tract was 40℃ and 30 ℃ respectively, and the optimum pH was 6.8 and 7.2 respectively. The optimum temperature of cellulase in hepatopancreas and digestive tract was 40~50℃ and 40℃ respectively, and the optimum pH was 5.2 and 4.8 respectively. The activities of amylase and cellulase in hepatopancreas are much stronger than those in digestive tract under the optimum temperature and pH range and have organic specificity, while there is no significant difference in the activities of pepsin and tryptase between hepatopancreas and digestive tract. The activities of different digestive enzymes are also significantly different in the same organ.
