长江中下游地区湖北钉螺线粒体CO I 基因遗传变异研究

从长江中、下游的江苏省丹徒区和邗江区、浙江省平湖市、湖北省武昌区、江西省余干县和彭泽县、安徽省安庆市和贵池区等5 个血吸虫病流行省8 个点现场采集湖北钉螺样本, 提取基因组DNA、PCR 特异性扩增线粒体DNA 细胞色素C 氧化酶亚单位1(CO I)基因, 用ClustalX 1.81 软件进行多序列比对, MEGA 4.0 软件中的Kimura 双参数法(Kimura 2-parameter)计算遗传距离, 邻接法(NJ)和最大简约法(MP)构建系统发生树。8 个地理株湖北钉螺(Oncomelania hupensis)共得到701 个同源位点,A/T(60.4%)含量高于C/G(39.6%), 所有序列间的平均遗传距离为0.025。进化树显示, 江苏丹徒、湖北武昌、江西余干及安徽安庆和贵池5 个地理株钉螺形成一个分支, 而浙江平湖、江苏邗江和江西彭泽3个地理株钉螺形成另一个分支; 但两种方法构建的系统发生树存在差异。长江中下游地区湖北钉螺线粒体CO I 基因遗传变异显著。

Genetic diversity of CO I in Oncomelania hupensis snails (Gastropoda: Rissooidea) along the middle and lower reaches of the Yangtze River
Abstract:
The Oncomelania hupensis snail samples were collected from eight locations including Dantu and Hanjiang of Jiangsu, Pinghu of Zhejiang, Wuchang of Hubei, Yugan and Pengze of Jiangxi, Anqing and Guichi of Anhui, in five schistosomiasis-endemic provinces along the middle and lower reaches of the Yangtze River. The genomic DNA of the snails were extracted, and CO I gene fragments were amplified by PCR, then purified and sequenced. Sequences were analyzed with software ClustalX Version 1.81. The genetic distances of the various regions were calculated using Kimura 2-parameter method and phylogenetic trees were constructed with NJ and MP methods in software MEGA Version 4.0. Following the editing of ClustalX, a total of 701 homologous sites were obtained, and the content of A/T (60.4%) was higher than C/G (39.6%). The mean genetic distance among all sequences was 0.025. The phylogenetic tree showed that the snails from Dantu of Jiangsu, Wuchang of Hubei, Yugan of Jiangxi, Anqing and Guichi of Anhui were clustered, and snails from other three sites developed another cluster. However, there was variation observed in the phylogenetic tree constructed by the two methods. It is indicated that there is distinct genetic diversity in CO I genes of O. hupensis snails along the middle and lower reaches of the Yangtze River in China.
