甲基睾丸酮在尼罗罗非鱼(Oreochromis niloticus)肌肉中的代谢和消除规律研究

摘要:
在实验水温(28±2)℃条件下, 以60mg/kg单剂量甲基睾丸酮对尼罗罗非鱼混饲口灌给药后, 采用超高效液相色谱-串联质谱联用(UPLC-MS/MS)法测定尼罗罗非鱼肌肉中的药物水平, 研究甲基睾丸酮在尼罗罗非鱼肌肉中的代谢及消除规律。结果表明, 肌肉中药物浓度时间数据符合一级吸收二室开放模型, 主要药代动力学参数达峰时间T(peak)为3.04h, 质量浓度C(max)为33.331ng/g, 消除半衰期T(1/2)β为23.073h。肌肉中甲基睾丸酮浓度24h内呈指数型下降, 具有较高的消除速率, 其后消除得较缓慢, 消除方程符合C= 46.18e?0.101t。29d后罗非鱼肌肉中甲基睾丸酮水平在美国FDA的检测限0.8ng/g以下, 给药120d后肌肉中检测不到甲基睾丸酮残留。

THE METABOLISM AND ELIMINATION OF 17α-METHYLTESTOSTERONE IN MUSCLE OF NILE TILAPIA OREOCHROMIS NILOTICUS

Abstract:
In order to study the metabolism and elimination of 17α-methyltestosterone in muscle of Nile tilapia, Oreochromis niloticus, single oral administration of 17α-methyltestosterone mixed in diet at 60mg/kg (BW) has been investigated in tilapia at (28±2)℃. The concentration of 17α-methyltestosterone in muscle of tilapia were determined by Ultro high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). The result demonstrated that concentration-time data of 17α-methyltestosterone in muscle was conformed to two-compartmental open model with first order absorption. The major pharmacokinetic parameters were as follows: T(peak)= 3.04h, C(max)= 33.331ng/g, T(1/2)β= 23.073h. 17α-methyltestosterone was eliminated exponentially, decreasing by 90% within 24h after feeding. Then it eliminated slower relatively. The elimination curve of 17α-methyltestosterone in muscle of tilapia was conformed to C= 46.18e?0.101t. After 29 days of feeding untreated diet, 17α-methyltestosterone represented under 0.8ng/g of muscle, which is the detection limit of FDA (Food and Drug Administration). 17α-methyltestosterone in muscle of tilapia could not detected 120d after cessation of 17α-methyltestosterone treated by UPLC-MS/MS.
