我国沿海鼠尾藻(Sargassum thunbergii)遗传资源 RAPD 分析

摘要:
采用 RAPD 分子标记技术分析了我国沿海五个海区(大连、威海、青岛、温州、福州)鼠尾藻种群的遗传多样性。 从 60 条随机引物中筛选出 21 条引物, 对五个鼠尾藻种群共 150 份 DNA 样本进行了扩增, 共检测到 207 个有效位点, 多态位点达 89.5%, Nei 基因多样性指数(H)为 0.2966, Shannon信息指数(I)为 0.4508; 种群间的遗传分化系数(Gst)为 0.6131, 基因流(Nm)为 0.3155。 AMOVA分析表明 Fst 为 0.6185。我国沿海鼠尾藻种群遗传多样性较低, 遗传分化水平较高。

POPULATION GENETIC DIVERSITY OF SARGASSUM THUNBERGII IN THE COAST OF CHINA

Abstract:
Five populations of S. thunbergii in Chinese coastal Dalian, Weihai, Qingdao, Wenzhou, and Fuzhou cities were analyzed to understand the genetic diversity of the species in Chinese coastal areas. Random amplified polymorphism of DNA (RAPD) analysis was conducted. Twenty one out of 60 polymorphic 10-base random primers were selected, from which 207 useful loci were detected. The polymorphic loci, Nei’s gene diversity and Shannon diversity index were: 89.5%, 0.2966, 0.4508, respectively. The coefficient of genetic differentiation was 0.6131 and the gene flow (Nm) was 0.3155. The results of AMOVA show significantly genetic differentiation among populations. Low level of genetic diversity within populations and high level of genetic differentiation among populations of S. thunbergii were revealed, which is important for the conservation of S. thunbergii germplasm and breeding.
