6种鳗鲡(Anguilla)线粒体DNA COⅡ基因序列分析及其系统发育

摘要:
采用基因测序的方法, 对 6 种鳗鲡的 COⅡ基因片段进行了 PCR 扩增和测序。结果表明, 6种鳗鲡 COⅡ基因片段的长度都为 412bp, 4 种碱基组成非常相似, 并且 A+T 的含量都大于 G+C 的含量。 6 种鳗鲡 COⅡ基因片段核苷酸序列之间有 46bp 的差异, 其中有 6 处发生碱基颠换, 40 处发生碱基转换。 非洲鳗鲡(Anguilla mossambic)和日本鳗鲡(A. japonica)COⅡ基因片段核苷酸序列差异最大为27bp, 同源性为 93.45%, 而日本鳗鲡和菲律宾鳗鲡(A. mormorata)COⅡ基因片段核苷酸序列差异最小为 5bp, 同源性为 98.79%。

Abstract:
MtDNA COⅡgene fragments of six Anguilla species were amplified using the PCR method, and the prod-ucts were purified and sequenced. The results showed that all the mtDNA COⅡgene fragments had 412bp with similar base composition, which had greater amount of the A+T pairs than the G+C pairs. The difference among the six mtDNA COⅡgenes was 46bp, including 6 base transversions and 40 base transitions. The difference between the mtDNA COⅡ genes of A. mossambica and A. japonica was 27bp with 93.45% homology; however the difference between those of A. japonica and A. mormorata was 5bp with 98.79% homology.
