两种培养温度下钝顶螺旋藻吸收累积锗的研究

摘要:
于1995年1–3月将钝顶螺旋藻培养在5–15mg/L锗(Ge)浓度的培养基内,研究两种培养温度下(25°C和30°C)藻细胞、蛋白质、脂类和碳水化合物结合锗的量,探讨温度影响螺旋藻吸收累积锗的机理。结果表明,在两种培养温度下钝顶螺旋藻均可吸收累积一定量的锗(22.44–52.13μg/g)并均匀地结合到蛋白质、脂类和碳水化合物中;藻细胞结合锗的量在低温下(25°C)随外加锗的浓度增加而增加,在高温下(30°C)与外加错的浓度关系不明显;两种培养温度下蛋白质、脂类和碳水化合物中结合锗的量均随外加锗的浓度增加而增加,但高温下(30°C)结合的锗量要高于低温下(25°C)的各相应值;低温下(25°C)三种大分子物质中锗的结合率较稳定(48%左右),高温下(30°C)则随外加错的浓度增加而明显增加(37.2%–72.0%)。根据实验结果推测,低温(25°C)主要影响螺旋藻对锗的吸附作用,高温(30°C)则影响锗与细胞内大分子物质的结合作用。

STUDY ON THE ACCUMULATION AND BINDING OF GERMANIUM IN SPIRULINA PLATENSIS CULTURED IN TWO TEMPERATURES REGIMES

Abstract:
The present experiment was carried out from January to March 1995; the algal species were provided by Laboratory of Diatom Research, Department of Biology, Xiamen University. Spirulina platensis grown in the Zarrouk's media with contents of dioxide germanium, i.e. 5.0, 10.0 and 15.0 mg/L germanium was cultured in two temperatures regimes (25°C and 30 °C). Absorption of germanium in algal cells and distribution of germanium in proteins, lipids and carbohydrates were studied. The results show that S. platensis can accumulate some amounts of germanium in cells (22.44–52.13 μg/g algal dry weight) and bind them with proteins, lipids and carbohydrates evenly in the two culture temperatures; the germanium absorbed by S. platensis increased with added germanium at the lower temperatures (25 °C), from 25.57 to 52.13 μg/g dry weight, but was not so sensitive to added germanium at the higher temperatures (30 °C), from 22.44 to 38.40 μg/g dry weight. The germanium bound to the proteins, lipids and carbohydrates increased with added germanium at the two temperatures, but the amount bound to these three macromolecular compounds at the higher temperature (30 °C) was higher than that at the lower temperatures (25 °C) was stable, about 48%. The ratio increased with added germanium at higher temperature (30 °C), from 37.2% to 72.0%. On the basis of the above results, lower temperatures mainly affects absorption of germanium, and higher temperature (30 °C) affects biochemical formation of germanium-macromolecular compounds with proteins, lipids and carbohydrates.
