条斑紫菜( Porphyra yezoensis) 耐高温品系的筛选及特性分析

摘要:
采用60Co-γ射线诱变和高温胁迫处理条斑紫菜野生品系(WT), 进行了耐高温品系的选育研究。结果表明, 在18℃和22℃组, 所选育的耐高温品系( TM-18 ) 和WT品系的壳孢子存活率无显著差异, 但在24℃和25℃组, TM-18 与WT相比, 壳孢子存活率分别提高了274%和296%, 畸形苗率分别下降了286%和161%, 假根发生率分别提高了151%和429%。将 18℃培养50d 的苗再分别在18℃、22℃和24℃下再培养30d, TM-18 苗的绝对生长率分别为WT苗的 3.82、3.16 和4.95 倍, 特定生长率为WT苗的1.53 、1.49 、2.10 倍。在24℃下培养15d 或25℃下培养10d, WT苗停止生长, 出现了卷曲和严重腐烂, 而TM-18 苗仍然生长良好, 不腐烂。另外, TM-18 的三种主要光合色素(Chl.a 、PE和PC) 含量明显比WT高。上述结果说明, TM-18 是一个颜色好、生长快和耐高温的优良品系。

SELECTION AND CHARACTERIZATION OF A HIGH-TEMPERATURE RESISTANT STRAIN OF PORPHYRA YEZOENSIS UEDA (BANGIALES, RHODOPHYTA)

Abstract:
A high-temperature-tolerant strain ( TM-18 ) of Porphyra yezoensis Ueda was isolated from the wild type ( WT) using by 60Co-γ -radiation and high-temperature cultivation. At 18 and 22℃, there was no significant difference on the sur-vival rates between the WT and TM-18 conchospores. However, at 24 and 25℃, the survival rates and the rhizoid germina-tion rates of conchospore-germlings of TM-18 were 274% and 296%, 151% and 429% higher than those of the WT, respectively. Furthermore, the occurrence rates of abnormal blades in TM-18 was reduced by 286% and 161% compared with the WT. When the 50-day-old conchospore-germlings initially cultured at 18 ℃ were cultured at 18, 22 and 24 ℃ for another 30 days, the specific growth rates of the TM-18 blades were 3.82, 3.16 and 4.95 times higher than the WT, respectively, and the absolute growth rates of the TM-18 blades were 1.53, 1.49 and 2.10 higher than the WT, respectively. The WT blades stopped growing and then severely decayed after being cultured for 15 days at 24℃ or 10 days at 25 ℃. In comparison, the TM-18 blades maintained normal growth. Finally, the contents of the three main photosynthetic pigments (Chl.a , PE and PC) in the TM-18 blades were significantly higher than those of the WT blades. This study showed that the TM-18 strain was superior to WT strain in growth, high-temperature tolerance, and pigment productions.
