中国沿海 7 个长蛸(Octopus variabilis)野生群体的遗传多样性

摘要:
基于线粒体 DNA 的 12S rRNA 和 COⅢ基因序列对我国沿海重要经济头足类长蛸不同群体(大连、烟台、青岛、连云港、舟山、温州、厦门)进行了遗传多样性和遗传结构的分析。由 PCR 扩增获得 80 个个体的 12S rRNA 基因 416bp、COⅢ基因 512bp 的部分序列, 两者多态性遗传参数统计显示, 80个个体分别检出 64 (12S)和 27 (COⅢ)个单倍型, 总群体单倍型多样性指数(Hd)分别为 0.984 (12S)和 0.909 (COⅢ), 核苷酸多样性指

GENETIC DIVERSITY OF SEVEN POPULATIONS OF OCTOPUS VARIABILIS IN CHINA’S COASTAL WATERS BASED ON THE 12S rRNA AND COⅢ GENE ANALYSIS

Abstract:
In this study, the genetic diversity and genetic structure of seven wild populations of Octopus variabilis in China’s coastal waters (Dalian, Yantai, Qingdao, Lianyungang, Zhoushan, Wenzhou and Xiamen) were analyzed using the mitochondrial gene sequencing technology (12S rRNA and COⅢ). The result revealed 64 haplotypes using the 12S rRNA gene and 27 haplotypes using the COⅢ gene for total 80 individuals obtained using PCR method. The haplotype diversities (Hd) were 0.9840 (12S) and 0.9092 (COⅢ), respectively; the nucleotide diversities (Pi) were 0.0277 (12S) and 0.0337 (COⅢ), respectively; and the average nucleotide differences (K) were 9.403 (12S) and 17.265 (COⅢ), respectively, showing high genetic diversity among populations. The genetic distances between the Xiamen population and other populations were up to 0.072 (12S) and 0.102 (COⅢ), respectively, showing considerable genetic differentiation, which is probably caused by different hydrological conditions in the Xiamen site than other sites.
