谷氨酰胺二肽对日本对虾血清生化指标、肝胰腺细胞凋亡及肠粘膜形态的影响

摘要:
采用饲养对比试验方法, 研究了谷氨酰胺二肽(Gln 二肽)对日本对虾血清生化指标、肝胰腺细胞凋亡和肠绒毛的影响及对虾体健康的作用。结果表明, 饲喂Gln 二肽各组的PO、LSZ、ACP 含量均显著(P<0.05)高于对照组, 添加0.5%和1.0% Gln 二肽组的SOD 和ALP 含量也显著(P<0.05)高于对照组; 添加1.0% Gln 二肽的TP 含量显著(P<0.05)高于其它各组。血清TP、PA、UN、CHO、PO、LSZ、SOD、ALP、ACP 和LDH 等各项指标均随Gln 二肽添加量的提高而呈现增加趋势。试验建立了流式细胞术检测对虾肝胰腺细胞凋亡率的方法; 肝胰腺细胞凋亡率随着日粮Gln 二肽添加量的增加而显著降低(P<0.05)。肠道切片显微观测结果表明, 添加1.0%的Gln 二肽能显著增加日本对虾肠绒毛高度(P<0.05)。饲料中添加Gln 二肽可显著提高日本对虾血清中的溶菌酶、抗氧化酶及磷酸酶活性, 降低肝胰腺细胞凋亡率, 增加肠绒毛高度, 改善虾体健康状况。

BIOCHEMICAL EFFECTS OF GLUTAMINE DIPEPTIDE ON PENAEUS JAPONICUS

Abstract:
The effects of glutamine dipeptide on serum biochemical parameters, wither of pancreas cells, morphology of intestinal mucosa in Penaeus japonicus are studied. 240 individuals in 9—10cm in length and 11—14g in body weight were randomly assigned to four treatments in three replicates and 20 prawns in each tank. The groups received in oral administration the same basal diet supplemented of 0%, 0.25%, 0.5% or 1.0% glutamine dipeptide, respectively. The trial lasted for 16 days. The results show that adding glutamine-dipeptide significantly increased serum phenoloxidase, lysozyme, and acid phosphatase activity (P<0.05). The serum superoxide dismutase and alkatine phosphatase activity was significantly higher in with 0.5% or 1.0% groups than the control (P<0.05), and significantly higher total protein concentration in plasma of the 1.0% group than the control and others (P<0.05). In addition, increases were also found in plasma total protein, prealbumin, uric nitrogen, cholesterol, phenoloxidase activity, lysozyme activity, superoxide dismutase activity, alkatine phosphatase activity, acid phosphatase activity, and lactate dehydrogenase. The apoptosis of the hepatopancreas cells was established in flow cytometry, showing that the apoptosis was significantly reduced (P<0.05); adding 1.0% glutamine dipeptide significantly increased the intestinal villi height (P<0.05). Therefore, glutamine dipeptide could enhance the activity of lysozyme, antioxidant enzyme, and phosphatase in plasma, decrease the rate of apoptosis of hepatopancreatic cells, and increase intestinal villi height in P. japonicus; in other words, the healthy of prawns were improved.
