中华乌塘鳢(Bostrichthys sinensis) 和大弹涂鱼( Boleophthalmus pectinirostris) 成熟产卵过程中17a-羟基孕酮和前列腺素水平的研究

摘要:
以酶联免疫吸附分析法测定了中华乌塘鳢和大弹涂鱼成熟产卵过程中卵巢液、精巢液、贮精囊液、雌鱼尿液和雄鱼尿液中的17a-羟基孕酮( 17a-P) 、前列腺素E2( PG E2) 和前列腺素F2a( PGF2a)的含量。结果表明, 中华乌塘鳢卵巢液中的17a-P和PGE2含量随着卵巢发育成熟而升高。性成熟前后卵巢液中的PGE2含量高于PGF2a。性成熟雄鱼尿液和贮精囊液中的PGE2含量显著高于精巢液或雌鱼尿液。性成熟前后雌鱼尿液中的17a-P、PGE2和PGF2a含量均显著高于卵巢液。性成熟和退化期雌鱼尿液中的PGE2含量明显高于PGF2a。性未成熟大弹涂鱼( 第Ⅳ期) 卵巢液中PGE2和PGF2a含量极低; 性成熟时( 第Ⅴ期) 卵巢液中的PGE2、PGF2a和17a-P含量显著上升, PGF2a含量显著高于PGE2

STUDIES ON 17a-P AND PGs LEVELS DURING MATURATION AND SPAWNING IN FISHES BOSTRICHTHYS SINENSIS AND BOLEOPHTHALMUS PECTINIROSTRIS

Abstract:
Levels of 17a-OH Progesterone ( 17a-P) , Prostaglandin E2( PGE2) and Prostaglandin F2a ( PGF2a) of ovarian extract, testis extract, seminal vesicle extract, female urine and male urine during maturation and spawning were determined using the Enzyme-linked Immunosorbent Assay ( ELISA) in the Chinese black sleeper ( Bostrichthys sinensis ) and the mudskipper (Boleophthalmus pectinirostris) . In the Chinese black sleeper, the results showed that levels of 17a-P and PGE2 increased as the ovary matured. The levels of 17a-P were in sequence order of female urine [ ( 35.48±6.20) ng/ml] > male urine [ (23.90±9.75)ng/ml] > testis extract [ ( 22.25±12.05) ng/ml] > seminal vesicle extract [ ( 20.38±8.94) ng/ml] > ovarian extract [ ( 4.98±3.30) ng/ml] at sexual maturation stage. The levels of 17a-P between female urine and ovarian extract were significantly different ( P< 0.01). The levels of PGE2 were higher than those of PGF2a in ovarian extract before and after maturation. The levels of PGE2 in mature male urine and seminal vesicle extract were significantly higher than those in mature female urine or in male testis extract. The levels of 17a- P, PGE2 and PGF2a in female urine were significantly higher than those in ovarian extract before and after maturation. The levels of PGE2 were obviously higher than those of PGF2a in female urine at both mature stage and during the degenerated stage. In the mudskipper, the levels of PGE2 and PGF2a of ovarian extract were extremely low in the mudskipper at immaturity stage Ⅳ. The levels of PGE2, PGF2a and 17a-P in ovarian extract increased significantly in females at maturity stage Ⅴ and the level of PGF2a was significantly higher than that of PGE2 ( P < 0.01). The Levels of 17a-P, PGE2 and PGF2a in ovarian extract were significantly higher than those in seminal vesicle extract or testis extract. Regarding prostaglandins, the levels of PGF2a were higher than those of PGE2 in ovarian extract, testis extract and seminal vesicle extracts. The results demonstrate that both PGE2 in Bostrichthys sinensis and PGF2a in Boleophthalmus pectinirostris are both specific prostaglandins.
