饵料微藻培育系统内海洋弧菌生长特点

摘要:
于1995年1–3月在中国科学院海洋研究所进行饵料微藻培育系统内海洋弧菌生长特点的研究。选取来自对虾及扇贝体内4株弧菌,研究其在4种饵料微藻培育系统内的生长特点,同时进行无菌海水内弧菌菌株生长对照实验。结果表明,与对照实验(弧菌菌株生长良好)相比,处于生长指数后期至静止期的微藻培育系统能够强烈排斥弧菌。处于生长初期的微藻培育系统排斥弧菌的能力相对较弱,随着藻细胞密度的增加而增强。长期培养的微藻培养系统,球等鞭金藻3011和扁藻1040由于老化严重,排斥弧菌能力相对减弱;而三角褐指藻2038及小球藻1061由于老化程度相对较弱,仍然具有强烈排斥弧菌能力。据此认为,饵料微藻培育系统可排斥弧菌。本文结果对采用生态调控防治疾病具有重要意义。

GROWTH FEATURE OF VIBRIOS IN MICROALGAE CULTURAL SYSTEMS

Abstract:
There are more and more diseases related to the marine aquacultured animals. Curative effects of the chemicals (include disinfectants, antibiotics and other substances) are unsatisfactory because these drugs can pollute environment and induce drug resistant bacteria. It is necessary to develop ecological controlling techniques to prevent the diseases to ensure the sustainable development of the marine aquaculture. Investigations were conducted into the effect of 4 marine unicellular algae cultural systems (mixtures of the algae and algae concomitant bacteria), Isochrysis galbana 3011 strain, Phaeodactylum tricornutum 2038 strain, Platymonas sp. 1040 strain and Chlorella sp. 1061 strain cultural systems on growth of 4 marine Vibrio spp, V. alginolyticus 21-1, V. anguillarm 65#, V. parahaemolyticus 10-1 and 87-1. Batch culture method was adopted. Control test of vibrios growth in sterilized seawater (0.2μm membrane filtered and autoclaved) was carried out simultaneously. Compared with control test (the vibrios grew well, all above 2.5×105 cfu/ml), cultural systems of these microalgae which are between the late exponential phase and the static phase of growth can effectively expel the vibrios. Densities of the vibrios decreased to 10 cfu/ml or lower from initially high (3.0×103 cfu/ml, respectively) within 24 hours (the 65# decreased to 10 cfu/ml within 48 hours in algae 3011 culture). Expelling efficiency of the cultural systems in the beginning phase of the growth weakens (because algal and algae concomitant bacterial densities decreased after the algae were transferred to new media), but it increased with algal growing (densities of the algal cell rise). The systems with long culture time (about one month) had less efficiency of restriction on growth of the vibrios when the algae (3011 and 1040, for example) aged obviously (cell sunk to bottom of the glass flasks and suspended algal cell densities decreased vehemently, algae 3011 cell density decreased from 4×106 cell/ml to 7.6×104 cell/ml, for example), but some cultural systems of algae (2038 and 1061) had high expelling efficiency because they had relatively high suspended algal cell densities. It seems that there is certain relationship between the suspended algal densities and expelling efficiency to vibrios. We suggest that the universal law that growth of vibrios are restricted by cultural system of microalgae may exist. Our results can provide theoretical foundation to prevent disease with ecological controlling technique.
