基于线粒体COI序列莱州湾单环刺螠遗传多样性分析

摘要:
单环刺螠(Urechis unicinctus)具有独特的分类学地位和广阔的经济前景。为系统地了解莱州湾单环刺螠遗传多样性,作者基于线粒体COI序列利用基因测序技术研究了莱州湾3个单环刺螠不同群体的遗传多样性。实验结果显示,84个莱州湾单环刺螠样本有变异位点数125个,其中包括43个单一变异位点, 82个简约信息位点;48个单倍型,单倍型多样性0.977;核苷酸多样性指数0.01048;平均核苷酸差异数量13.725;莱州湾单环刺螠遗传多样性水平较高;个体聚类并没有体现出和地理位置相关。本研究结果为莱州湾单环刺螠的种质资源保护与开发提供了参考依据。

Analysis of genetic diversity of Urechis unicinctusin in Laizhou Bay based on the mitochondrial COI gene sequence

Abstract:
Urechis uniconctus has a unique taxonomic status and a broad economic prospect. The mitochondrial COI sequence were detected in Urechis uniconctus from three populations in Laizhou Bay of Shandong province to analyze the genetic diversity of Urechis uniconctus. And 70 pieces of fragments with the length of 1309bp were obtained after treatment. Results showed that, there were 115 mutation sites in Urechis uniconctus in Laizhou Bay of Shandong province, including 43 single mutation loci, 82 minimalism information loci, 77 double mutation minimalism information loci, and 5 triple mutation loci. The diversity of haplotypes was 0.977. Nucleotide diversity index was 0.01048. The mean difference number of nucleotides was 13.725. Urechis uniconctus in Laizhou Bay of Shandong province has a high genetic diversity. The results of this study provide basic information for the conservation and development of Urechis uniconctus in Laizhou Bay of Shandong province.
