金属蛋白酶对大菱鲆血清抗氧化酶活性及丙二醛含量的影响

摘要:
为了研究金属蛋白酶对大菱鲆(Scophthalmus maximus)抗氧化系统的影响,分别对大菱鲆肌肉注射不同浓度的金属蛋白酶并于3、6、12、24、48、72 h测定血清中谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、总超氧化物歧化酶(T-SOD)、总抗氧化活力(T-AOC)以及丙二醛(MDA)含量变化。结果显示,低浓度组(0.037 mg/mL)金属蛋白酶处理后,CAT、GSH-Px活力除3 h显著高于对照组外(P<0.05),其余时间点均与对照组无显著差异(P>0.05),而低浓度组T-SOD和T-AOC在整个试验过程中均与对照组无显著性差异(P>0.05)。中浓度组(0.073 mg/mL)金属蛋白酶胁迫后血清中CAT、GSH-Px、T-SOD和T-AOC在3-72 h均呈现波浪形的变化趋势。高浓度组(0.147 mg/mL)金属蛋白酶对上述4种抗氧化酶产生不同程度的抑制,且在6-72 h均显著低于对照组(P<0.05)。金属蛋白酶低浓度和中浓度组MDA含量呈现较为平稳的变化趋势,且与对照组无显著性差异(P>0.05),而高浓度组MDA含量在3-72 h均显著高于对照组(P<0.05)。

Effects of Metalloprotease on antioxidant system enzyme activities and malondialdehyde content of serum of Scophthalmus maximus

Abstract:
A metalloprotease was purified from the extracellular products (ECP) of Vibrio anguillarum. It was lethal to S. maximus when injected intramuscularly. However, there is little information about the effects of metalloprotease on antioxidant system and lipid peroxidation in S. maximus. In this study, we measured catalase (CAT), glutathione peroxidase GSH-Px) total antioxidation activity (T-AOC), total superoxide dismutase (T-SOD) activity and malondialdehyde (MDA) content in the serum exposed to 0.037 mg/mL, 0.073 mg/mL or 0.147 mg/mL metalloprotease, respectively, at 3, 6,12, 24, 48, 72 hours. The results showed that: compared to the control group, the activities of CAT and GSH-Px were no significant difference at test process except at 3 h (P> 0.05). The variation of T-SOD and T-AOC was not obvious in serum of low concentration group. The changes of CAT, GSH-Px, T-SOD and T-AOC in the serum of the medium concentration group after metalloproteinase stress were wavy at 3-72 h. Metalloproteinase in the high concentration group was inhibited in the above four antioxidant enzymes and were significantly lower than the control group at 6-72 h. The MDA content of low and medium concentration group was no significant different at 3-72 h (P> 0.05), while the MDA content of high concentration group was significantly higher than that of the control group at 3-72 h (P < 0.05).
